here's a few things off the top of my head:

The French Quarter: history+culture+architecture; nothing else like it in North America; just stroll around and soak it in!

FOOD: be sure to try the Gumbo, Jambalaya, shrimp etouffee, Poboy sandwhiches, etc...

Restaurants:

-Mother's (corner of Poydras and Tchoupitoulas in the Central Business District, close to French Quarter); casual old skool joint; during high tourist season there are line ups out the door of peeps waiting to be seated.

-Acme Oyster House (on Iberyville, between Bourbon and Royal); fresh oysters shucked in front of u and served on the half shell; splash it with tabasco sauce and a dash of horseradish and you're good to go!

-Cafe Du Monde (Decateur+St Anne); open since 1860, 24 hrs/day; must get the chicory-laced cafe au lait and beignets: puffy pillows of dough which are lightly fried and then doused with powdered sugar!!!!!

Bourbon St:

- if you prefer your beers cheap and you get a big kick out of drinking out on the street, then hit it up! Worth checking out.

Graveyard:

-bodies buried above ground in the most elaborate tombstones you'll ever see! many spots to choose from but the closest to the French Quarter is on the corner of Basin+St Louis;

Frenchmen St: bohemian part of town where there are several Jazz clubs to pick from

i concur with every one of tenenbaum's suggestions! nailed the highlights!

in addition, check out the homes in the Garden District. Some are mansions that are stunningly beautiful - close to Tipatina's too, if you can manage to get to a late night show there. also, see what's on at the Howlin' Wolf. the House of Blues is cool, you can go to just eat if no shows, but be prepared for the most packed club you will ever see if there's a good show there.

and drinking is encouraged. when you leave a club, they pour your drink into a plastic cup which you are free to take anywhere. the ONLY rule is no glass bottles while wandering.
